چکیده

Drugs. The Drug Synthesis and Chemistry Branch, Division of Cancer Treatment, NCI, National Institutes of Health (NIH), provided camptothecin (CPT) and flavopiridol (FLV). Aphidicolin (APD), 5,6-dichlorobenzimidazole 1-β-D-ribofuranoside (DRB), and α-amanitin were obtained from Sigma (St Louis, MO). Hydrogen peroxide (H2O2) was obtained from Fisher Scientific (Pittsburgh, PA). Iodouridine (IdU) was obtained from Sigma. The ATM kinase inhibitor (KU55933) (Hickson et al, 2004) and the DNA-PK kinase inhibitor (KU57788) (Leahy et al, 2004) were obtained from Kudos Pharmaceuticals (Cambridge, UK).

Etoposide Induces ATM-Dependent Mitochondrial Biogenesis through AMPK Activation

BACKGROUND DNA damage such as double-stranded DNA breaks (DSBs) has been reported to stimulate mitochondrial biogenesis. However, the underlying mechanism is poorly understood. The major player in response to DSBs is ATM (ataxia telangiectasia mutated). Upon sensing DSBs, ATM is activated through autophosphorylation and phosphorylates a number of substrates for DNA repair, cell cycle regulation...

Ataxia telangiectasia mutated activation by transcription- and topoisomerase I-induced DNA double-strand breaks.

Ataxia telangiectasia mutated (ATM), the deficiency of which causes a severe neurodegenerative disease, is a crucial mediator for the DNA damage response (DDR). As neurons have high rates of transcription that require topoisomerase I (TOP1), we investigated whether TOP1 cleavage complexes (TOP1cc)-which are potent transcription-blocking lesions-also produce transcription-dependent DNA double-st...

Transcription-coupled DNA double-strand breaks are mediated via the nucleotide excision repair and the Mre11-Rad50-Nbs1 complex.

The cellular activity of Yondelis (trabectedin, Ecteinascidin 743, Et743) is known to depend on transcription-coupled nucleotide excision repair (TCR). However, the subsequent cellular effects of Et743 are not fully understood. Here we show that Et743 induces both transcription- and replication-coupled DNA double-strand breaks (DSBs) that are detectible by neutral COMET assay and as gamma-H2AX ...

Activation of ATM and histone H2AX phosphorylation induced by mitoxantrone but not by topotecan is prevented by the antioxidant N-acetyl-L-cysteine.

The DNA topoisomerase II (topo2) inhibitor mitoxantrone (MXT) and topo1 inhibitor topotecan (TP) are antitumor drugs widely used to treat different types of cancer. Their mechanism of action is thought to stabilize otherwise transient ("cleavable") complexes between topo2 or topo1 and DNA; the collisions of the DNA replication fork during replication, or RNA polymerase during transcription, wit...

γ-H2AX as a protein biomarker for radiation exposure response in ductal carcinoma breast tumors: Experimental evidence and literature review

Background: H2AX is a histone variant that is systematically found and ubiquitously distributed throughout the genome. DNA double-strand breaks (DSBs) induce phosphorylation of H2AX at serine 139 (γH2AX), an immunocytochemical assay with antibodies recognizing γH2AX has become the gold standard for the detection of DSBs. The importance of this assay to investigate different individu...

Molecular Pathways Transcription-Induced DNA Double Strand Breaks: Both Oncogenic Force and Potential Therapeutic Target?

An emerging model of transcriptional activation suggests that induction of transcriptional programs, for instance by stimulating prostate or breast cells with androgens or estrogens, respectively, involves the formation of DNA damage, including DNA double strand breaks (DSB), recruitment of DSB repair proteins, and movement of newly activated genes to transcription hubs. The DSB can be mediated...
